The FirAlgos study finds that effervescent oxycodone/paracetamol delivers early pain relief while improving daily functioning and patient-perceived treatment response in adults with moderate-to-severe pain.
A real-world study, led by R Vellucci et al., examined whether an effervescent formulation of oxycodone/paracetamol could offer a practical alternative for patients requiring analgesia, particularly those who may have difficulty swallowing conventional oral medicines. The FirAlgos study evaluated treatment outcomes in 94 adults treated at a tertiary pain therapy centre between 2022 and 2023. Participants had a mean age of 69.3 years, and most (75.5%) were being treated for non-oncologic pain.
Pain reduction was evident within the first week. Numeric rating scale (NRS) scores decreased significantly by day 7 (p<0.05) across pain dimensions in both oncologic and non-oncologic pain groups, with improvements maintained at Day 14. Patient-reported outcomes followed a similar pattern, with Patient Global Impression of Change (PGIC) scores shifting toward greater perceived improvement.
Importantly, the benefit extended beyond pain intensity. Brief Pain Inventory (BPI) assessments showed improvements in general activity, mobility, and work capability, suggesting that the observed response was accompanied by better functional performance. The findings were encouraging but were interpreted within the study's design. As a retrospective observational analysis without a comparator group, the study did not establish comparative efficacy or demonstrate that the effervescent formulation was superior to other analgesic options.
The relatively small sample and 14-day follow-up also limited broader conclusions. Overall, the FirAlgos findings provided real-world evidence on pain relief, functional outcomes, and patient-perceived response with effervescent oxycodone/paracetamol and supported the need for controlled studies to further evaluate its clinical effectiveness and acceptability.
