Background

Polycystic ovary syndrome (PCOS) is a common endocrine disorder frequently linked with menstrual irregularity, anovulation, and hormonal disturbances. Since impaired ovulation is a major contributor to reproductive difficulties in PCOS, strategies that support restoration of ovulatory function are clinically relevant. This study explored whether myo-inositol supplementation could improve ovulation and menstrual regularity in PCOS.

Method

In this prospective interventional study, 72 women with PCOS received an oral regimen of myo-inositol at a dose of 2000 mg twice daily, co-administered with folic acid, over a 12-week interventional period. Comprehensive clinical, biochemical, and transvaginal ultrasound parameters were documented at baseline and post-treatment.

Ovulation confirmation was performed via mid-luteal serum progesterone quantification or serial ultrasound follicle monitoring. Statistical evaluations comparing pre- and post-treatment outcomes were carried out via paired t-tests for continuous variables and chi-square tests for categorical parameters.

Result

Following 12 weeks of myo-inositol administration, 58.3% (n = 42/72) of women successfully attained ovulation, and 65.3% (n = 47/72) attained regular menstrual cycles. Remarkable reductions were noted in both the luteinizing hormone/follicle-stimulating hormone (LH/FSH) ratio and serum testosterone levels. The therapeutic protocol was well-tolerated across all participants, with zero major adverse events recorded.

Conclusion

Myo-inositol supplementation successfully restored ovulation, improved menstrual regularity, and normalized key hormonal markers in women with PCOS. Given its excellent safety profile, it serves as a potent, non-hormonal alternative for ovulation induction in clinical practice.
